Characteristics of serum separation gel

1. Thixotropy 

The molecular structure of serum separation gel contains a large number of hydrogen bonds, which form a three-dimensional network structure through hydrogen bonding. Under the action of centrifugal force, the network structure is destroyed and the colloid becomes a low viscosity fluid; After centrifugation, the hydrogen bond is reconnected to restore the high viscosity gel state, realizing the reversible change of dynamic structure. 

2. Density adaptability 

Its density is designed to be between 1.04-1.05 g/mL, which is between serum (1.026-1.031 g/mL) and blood cells (1.090-1.095 g/mL). After centrifugation, a stable isolation layer can be formed between serum and blood cells, achieving efficient separation. 

3. Physiological inertia 

Serum separation gel is chemically inert, insoluble in water, and has no biological activity, avoiding reaction with blood components and ensuring the stability of the test sample. For example, it does not interfere with clinical biochemistry and immune testing projects. 

4. Isolation performance 

The formed isolation layer can effectively prevent substance exchange between blood cells and serum/plasma (such as potassium ion leakage inside red blood cells), reduce hemolysis and fibrin interference, and maintain sample composition stability for up to 8 days (under plastic tube material). 

5. Material adaptability 

Plastic tube: The separation glue adheres tightly to the hydrophobic plastic tube wall, forming a complete isolation barrier and prolonging the storage time of serum. 

Glass tube: Due to its hydrophilic nature, it is easy to form a water molecule adsorption layer, resulting in weak isolation effect, and the serum storage period is only 2-3 days. 

6. Application compatibility 

Suitable for various blood collection tubes, such as SST coagulation tubes (to accelerate serum separation), EDTA nucleic acid tubes (to block hemoglobin interference), CPT monocyte preparation tubes (to separate lymphocytes), etc., to meet different detection needs. 

7. Synergistic promotion of coagulation 

Serum separation gel is often used in combination with coagulants to accelerate serum separation and improve detection efficiency by shortening blood clotting time (about 30 minutes). The coagulant is pre coated on the inner wall of the test tube in the form of anhydrous ethanol suspension, and its synergistic effect with the separation gel can reduce the risk of hemolysis. 

8. Sensitivity to centrifugal conditions 

Its performance depends on specific centrifugal parameters (such as a conventional centrifugal speed of 3000-3500 rpm for 10-15 minutes). Insufficient centrifugal force can cause the separation gel to fail to completely flip and form an isolation layer, leading to hemolysis or mixing of serum/blood cells, which affects the accuracy of detection. 

9. Anti interference stability 

High quality separation gel can effectively block fibrinogen interference and prevent fibrin filaments from blocking the detection instrument in serum after centrifugation. This characteristic is closely related to the colloid purity and production process. 

10. Temperature tolerance 

Maintaining stability at regular laboratory temperatures (18-25 ℃), extreme low temperatures (<4 ℃) may cause changes in the physical form of colloids, but they are reversible after returning to room temperature and do not affect isolation function. 

11. Biocompatibility 

It does not contain endotoxins or animal derived components, avoiding activation of the complement system or interference with immune detection results. It is suitable for high-sensitivity detection techniques such as enzyme-linked immunosorbent assay and chemiluminescence.
